چکیده

Highly purified isolates of potato spindle tuber viroid (PSTV) from Lycopersicon esculentum cv. Rutgers and citrus exocortis viroid (CEV) from Gynura aurantiaca have been compared. Two-dimensional fingerprinting analysis of PSTVand CEV-RNA labeled in vitro with lzrI has demonstrated that (i) each of these RNA species has a complexity compatible with the size estimate of 250X350 nucleotides and (ii) these RNA molecules do not have the same sequence. Polyacrylamide-gel electrophoretic studies suggest, among other things, that there could be some sequence and/or structure heterogeneity within each of these viroid RNA’s,

Viroids and their potential danger to potatoes in hot climates

Viroids are the smallest agents known to be pathogenic to plants. Unlike viruses, they are devoid of a protective protein coat and are composed entirely of circular ribonucleic acid of low molecular weight (ca. 85,000-130,000 d. In spite of their small size, viroids cause serious diseases of avocado, chrysanthemum, citrus, coconut, cucumber, hop, potato and tomato. Although viroid diseases have...

Viroids are autonomously replicating, small singlestranded circular RNA molecules that do not code for proteins and may cause disease in infected, susceptible plants. Viroids have the ability to induce both RNA-mediated transcriptional gene silencing (TGS) and posttranscriptional gene silencing (PTGS) in infected plants. Viroid PTGS has also been demonstrated in a wheat germ extract system. A p...

Nudeotkle Sequence of Cucumber Pale Fruit Virotd: Homology to Hop Stunt Viroid

Double stranded cDNA of cucumber pale fruit viroid (CPFV) has been cloned by the method of Okayama and Berg (Mol.Cell.Biol.2,161-170 (1982)) and the complete nucleotide sequence was established. The covalently closed circular molecules of single-stranded CPFV RNA consists of 303 nucleotides. The nucleotide sequence of CPFV was compared with the previously established sequence of hop stunt viroi...

Virp1 is a host protein with a major role in Potato spindle tuber viroid infection in Nicotiana plants.

Viroids are small, circular, single-stranded RNA molecules that, while not coding for any protein, cause several plant diseases. Viroids rely for their infectious cycle on host proteins, most of which are likely to be involved in endogenous RNA-mediated phenomena. Therefore, characterization of host factors interacting with the viroid may contribute to the elucidation of RNA-related pathways of...

Structural differences within the loop E motif imply alternative mechanisms of viroid processing.

Viroids replicate via a rolling circle mechanism, and cleavage/ligation requires extensive rearrangement of the highly base-paired native structure. For Potato spindle tuber viroid (PSTVd), the switch from cleavage to ligation is driven by the change from a multibranched tetraloop structure to a loop E conformation. Here we present evidence that processing of Citrus viroid III (CVd-III), a memb...
